What Does Yeast Look Like Under A Microscope

Place a coverslip on top and observe under different magnifications.
A single yeast cell is typically about 3 to 4 micrometers in diameter but there are some species that can grow to be much larger around 40 micrometers. A sample of the vaginal discharge is placed on a glass slide and mixed with a salt solution. Place a drop of the yeast mixture on the microscope slide it might be necessary to dilute it a bit more with water. The next layer of the cell wall is present on the outside of the cell that appears transparent.
Various stages of bud development are seen. There is nothing special about the appearance of the discharge itself that can make it clear that the discharge is due to yeast. It is also possible to observe the buds which can be seen on some of the yeast cells. They are units of protoplasm that contain nucleus globules of oil vacuoles and are enclosed by a cell wall.
The diagnosis is often made more clear by doing a special stain called a koh stain on the discharge and looking at it under the microscope. When we look at the yeast under the high power microscope we will see that it has numerous microscopic bodies that are shaped like an egg. High magnifications will be needed to see the yeast well. The large spherical area is the nucleus while the granulated part is the cytoplasm of the cell.
